General description

Histone-lysine N-methyltransferase NSD3, also known as Nuclear SET domain-containing protein 3, Protein whistle, WHSC1-like 1 isoform 9 with methyltransferase activity to lysine or Wolf-Hirschhorn syndrome candidate 1-like protein 1, (WHSC1-like protein 1), and encoded by the gene WHSC1L1/NSD3, is a histone methylase that preferentially methylates Lys-4 and Lys-27 of histone H3. Lys-4 methylation is generally a specific tag for transcriptional activation while Lys-27 methylation is a mark for transcriptional repression. NSD3 is highly expressed in many tissues including the brain, heart and skeletal muscle. Overexpression of NSD3 is associated with certain forms of lung cancer as well as certain other cancers. NSD3 also facilitates the expression of many other genes important in cancer progression such as Brd4. Brd4 through its extraterminal domain interacts with NSD3 which regulates transcription by modifying the chromatin microenvironment at Brd4 target genes.

Target description

~180 kDa, ~160 kDa, ~90 kDa, and ~70 kDa observed. Uniprot desribes four isoforms at 162 kDa (isoform 1), 156 kDa (isoform 2), 73 kDa (isoform 3), and isoform 4 (155 kDa). This protein is known to be acetylated and phosphorylated and may be observed at higher molecular weights. Uncharacterized band(s) may be observed in some cell lysates.
